- The purpose of DNA Extraction is to get pure DNA out of your sample.
- In this video, you will learn how to use an extraction kit. This kit is for soil.
- The kit contains the chemicals and supplies you need to extract DNA.
- The first time you use the kit, add ethanol to the concentrated buffers.
- Be sure to mark the tops of the buffer bottles.
- Before starting, if a heat block is needed, turn it on and set it to the correct temperature.
- Label the tops of the spin columns and microcentrifuge tubes.
- The first step is LYSIS. This breaks open the cell to free the DNA.
- If extracting DNA from tissue, add Buffer ATL and proteinase K. Follow specific directions if extracting from plants or soil.
- Vortex the sample to mix.
- Place in heating block until tissue is completely dissolved. Vortex occasionally.
- When sample is completely lysed, vortex for 15 seconds.
- Add first Wash Buffer. The wash buffers precipitate DNA and rinse away proteins.
- Pipet into a labeled spin column labeled. Spin columns have a filter that binds DNA.
- Place samples in centrifuge and secure cover before closing lid.
- Centrifuge samples for recommended speed and time.
- Discard the flow-through and old collection tube. Place spin column in a new collection tube.
- Add second wash buffer.
- Centrifuge samples for recommended speed and time.
- Place column in a clean, LABELED 1.5ml microcentrifuge tube. Pipet Elution Buffer or water directly onto the DNEasy membrane.
- Incubate for 1 minute to extract DNA.
- Centrifuge samples for recommended speed and time.
